Choristoneura fumiferana granulovirus (ChfuGV), member of Baculoviridae , is considered a very attractive and powerful candidate to control spruce budworm’s population, the most destructive pest of coniferous trees in North America. The cycle of GV’s infection involves a number of viral proteins, which for most of them have not been characterized. Among these proteins, baculovirus IAPs (Inhibitor of Apoptosis Proteins) seem to prevent the activation of caspases which are critical effectors of programmed cell death, a crucial process for host development and tissue homeostasis. This work aims to provide the IAP molecular characterization of ChfuGV. The gene coding for IAP has to be identified and amplified by PCR, cloned in pCR2.1 vector and then sequenced. The IAP amino acid sequence derived from the nucleotide sequence of the gene will be compared to that of other baculoviruses related phylogenetically. Also, the secondary and tertiary structure of the protein will be studied by computer analysis. The identification of ChfuGV’s proteins will lead to a better comprehension of granulovirus and insect interaction. |
