Enhancement of phagocytosis and survival of human neutrophils by PBI-1402: Potential protection against opportunistic infections.


Abstract Text
PBI-1402 is a low molecular weight synthetic molecule which has found use in some pharmaceutical preparations. In man, this compound is devoid of side effects at doses greater than 1 g/kg. Recently, we demonstrated that PBI-1402 is a potent activator of neutrophils with in vitro and in vivo activity comparable to Granulocyte Macrophage-Colony Stimulating Factor (GM-CSF). PBI-1402 enhances the phagocytic activity of human neutrophils. An enhancement of two to three times relative to control is observed depending on the immune status of the donor. Neutrophils have also a short half-life (6 hours in blood) and rapidly undergo characteristic changes indicative of apoptosis. Neutrophils obtained from healthy volunteers were assessed for apoptosis in presence or absence of PBI-1402 using the annexin V-FITC/PI method. After 24-hour incubation, PBI-1402 enhances human neutrophil survival by up to 90%. Further, PBI-1402 rescues doxorubicin-induced apoptosis of neutrophils. A protection of up to 90% is observed at concentration of up to 12 mM. In view of the importance of neutrophils in immune defense against microorganisms, PBI-1402 is targeted as an adjunct to cancer chemotherapy, bone marrow transplantation and diseases involving neutropenia.
