The Montreal Biodome operates a methanol-fed denitrification system that removes nitrate from its 3 million liters seawater tank. A bacterial isolate, named strain NL21, was isolated from this system. This organism is a facultative gram-negative rods-shaped (1 µm x 3 µm) bacterium. The optimum pH and optimum temperature for growth is 7-7,5 and 35°C, respectively. Phylogenetic analysis of the 16S rDNA sequence revealed that strain NL21 is a new lineage in the family of Phyllobacteriaceae within the alpha subgroup of Proteobacteria. Closest related genera are the Aminobacter, Pseudaminobacter, Mesorhizobium and Defluvibacter. The major cellular fatty acids are 18:1 w7c (75%), 19:0 cyclopropane w8c (9,4%) and 18:0 (4,2%). The G+C content is 57 mol/% that is different from the others Phyllobacteriaceae (61-63 mol/%). Strain NL21 reduces nitrate to nitrite but does not reduce nitrite to nitrogen gas. Only few sugars and amino acids can serve as carbon sources. Strain NL21 can grow without salt but addition of 1% NaCl stimulate its growth, and is tolerant up to 5% NaCl. We propose that strain NL21 is a new species that belongs to a new genus in the family of Phyllobacteriaceae. |
